Cucumber Mosaic Virus and Pepper Mild Mottle Virus in Pepper Growing Areas in Burdur Province, Turkey

Abstract


This study was carried out to detect the presence Cucumber mosaic virus (CMV) and Pepper mild mottle virus (PMMoV) in the pepper growing areas in Burdur province. A total of 124 plant samples were collected from pepper plantations with virus like symptoms in 2012-2013. Identification of viruses was performed using serological and molecular methods. As a result of DAS-ELISA (Double-antibody sandwich enzyme linked immunosorbent assay) tests, it was observed that 20 samples out of 124 were infected with viruses. CMV in 8 samples, PMMoV in 8 samples were found to be present. It was seen that 4 samples were infected with two viruses. RT-PCR (Reverse transcription polymerase chain reaction) was carried out by using specific primer which amplified a 678 bp fragment of coat protein of CMV in samples. Thus, CMV product of expected size in leaf samples were observed.  On the other hand, the expected amplification bands in 830 bp were not observed for PMMoV. The present study provides evidence for occurrence of CMV and PMMoV in Burdur province in Turkey based on serological methods. RT-PCR studies confirmed DAS-ELISA results for the detection of CMV.
